KennenHutchison FacultyMentor:CatherineMiller-Hunt Biology Chikungunyavirus(CHIKV)isaflavivirusthatistransmittedbyAedesaegyptiandAedesalbopictus mosquitoes.Althoughthevirushasalowmortalityrateinhumans,itcausesseveralseveresymptoms. In2005,anoutbreakofCHIKVwasidentifiedontheFrenchLaReunionIslandandinfectedmorethan 75%oftheisland’spopulation.ThisoutbreakwaslaterattributedtoaCHIKVstrainthatexhibiteda singleaminoacidchangeintheviralglycoprotein,andusedtheAe.albopictusmosquitoasavector.The mutation,analaninetovalinechangeataminoacid226(A226V),allowedforaleapinvector,from primarilyAe.aegyptitoprimarilyAe.AlbopictusandresultedinahighnumberofCHIKVcasesonthe Island.ThefactthatoneaminomutationcanexpandvectorcompetencythroughoutAedesspeciesisof interesttoourlab.TheaimofourprojectistoidentifyifotheraminoacidmutationsinCHIKV’s glycoproteincouldpermitorincreaseCHIKVcompetencyinAedesspeciescurrentlylocatedinwestcentralIllinois.Atthistime,invasiveAedesspeciesalbopictus,japonicusand.triserriatusarefound throughoutIllinois.Theresultsofourstudywillhelptoindicateifwest-centralIllinoiscurrentlyhasa mosquitovectorthatiscompetentforCHIKVinfectionandcouldthereforeputthecommunitiesof west-centralIllinoisatriskoffacingthevirusbeforetheinvasionofAe.aegyptireachesourstate.Asa secondaryobjective,ourresearchwilllookatthemechanismsofviralentrybyCHIKVintomosquito cells.
