LAPORAN INISIATIF INOVASI (KEMENTERIAN SUMBER ASLI & ALAM SEKITAR / INSTITUT PENYELIDIKAN PERHUTANAN MALAYSIA) Nama Inovasi : Ringkasan Inovasi Tujuan Inovasi Elemen Keberkesanan Development of protocol for the production of Acacia hybrid synthetic seeds The axillary buds of in vitro Acacia hybrid were cut aseptically where the explants were placed in the encapsulation matrix (sodium alginate solution). The encapsulated explants or synthetic seeds or beads were cultured onto the tissue culture medium and maintained in the culture room at 261°C under a 16 hour photoperiod with a photon flux density of about 35 molm-2s-1 provided by cool white fluorescent lamps to observe the germination ability of the beads. The beads are regarded as germinated when the expanded leaves appeared out from the capsules. Plantlets formation was observed once the shoots elongated and there were occasions where roots began to emerge. The synthetic seeds of Acacia hybrid cultured in vitro sprouted well into seedlings under suitable conditions. With the ability to germinate easily, the synthetic seeds production showed that the main advantage of the system is the combination of high-volume production and low-cost propagation. Synthetic seeds of Acacia hybrid can be used as an alternative planting material meant for forestry sector in the future and for germplasm conservation. This technique will become the alternative to high-cost vegetative propagation technologies. The use of synthetic seeds will become popular for green house production. The synthetic seeds of Acacia hybrid have the potential to be used as a substitute when plants fail to produce seeds naturally, or when they produce only a small number of fertile seeds. Direct sowing is possible with the seed sized propagules, thus eliminating the acclimatization step normally required in transplanting of mericlones. Synthetic seeds of Acacia hybrid has a potential in providing an inexpensive plant delivery system. The process of planting can be easily mechanised and this allows the direct delivery of tissue cultured plant propagules to the field. It also provides rapid bulking up for the production of individual genetically engineered plants especially for the highly demanded species. This may be a reliable method for long-term storage of plant genetic resources without apparent risk of genetic instability using minimum space and with lower labour and maintenance costs.
