FLUID THIOGLYCOLLATE MEDIUM w/ 0.5% BEEF EXTRACT BASE INTENDED USE Remel Fluid Thioglycollate Medium w/ 0.5% Beef Extract Base is a medium recommended for use in qualitative procedures to test for bacteria in biological products. SUMMARY AND EXPLANATION 1 Thioglycollate Medium was first described by Brewer in 1940. This medium contained a small amount of agar and a reducing substance to promote the growth of anaerobes. Fluid Thioglycollate Medium w/ 0.5% Beef Extract is recommended by the Animal and Plant Health Inspection 2 Service, U.S. Department of Agriculture (USDA). PRINCIPLE Sodium thioglycollate is a reducing agent which maintains a low oxygen tension in anaerobic media. A small amount of agar is added to impede diffusion of oxygen. Casein peptone and cystine supply nitrogenous and carbon compounds, while the sodium chloride maintains osmotic equilibrium. Beef extract provides nitrogenous substances and enhances organism recovery. Resazurin is added as an oxidationreduction indicator which turns pink when increased oxidation occurs. Yeast extract provides B-complex vitamins to enhance the growth of bacteria. Suspend 34.5 g of medium in 1000 ml of demineralized water. Heat to boiling with agitation to completely dissolve. Dispense into appropriate containers. Sterilize by autoclaving at 121°C for 15 minutes or following established laboratory procedures. PROCEDURE 1. Consult current editions of appropriate references for the recommended procedure for sample preparation, inoculation, testing, and interpretation. QUALITY CONTROL Each lot number of Fluid Thioglycollate Medium w/ 0.5% Beef Extract Base has been manufactured, packaged, and processed in accordance with current Good Manufacturing Practice regulations. All lot numbers have been tested using the following quality control organisms and have been found to be acceptable. Testing of control organisms should be performed in accordance with established laboratory quality control procedures. If aberrant quality control results are noted, sample results should not be reported. A slight turbidity (haziness) may be present due to the small amount of agar in the medium. Do not boil more than once; frequent boiling results in the development of toxic products.
