residue
is
increasingly recognised as a key phosphorylation event. Proteins containing
phosphohistidine (pHis) are implicated in various mammalian cellular processes
including regulation of ion channels, apoptosis, cell proliferation and differentiation,
inflammation, chromatin biology, cancer and cell signalling. However, much remains
to be understood about the role and extent of the modification in mammalian cell
biology. The study of both the extent and functional role of pHis in signaling, either
in vitro or in vivo, is complicated by the instability of the pHis modification. As a
consequence, we are currently handicapped by a chronic lack of chemical and
biochemical tools with which to study histidine phosphorylation. The aim of this
project is to develop tools for the study of the role of histidine phosphorylation in
mammalian cells. We will incorporate novel stable analogues of phosphohistidine
that we have recently synthesized into relevant peptide sequence to develop potent
and specific reagents as protein-protein interaction and PHP inhibitors.

research experience.
