1) Describe three ways in which bacteria can acquire DNA.
2) How do plasmids move between different bacteria? Can plasmids move between
different species? Different genera?
3) What is the fate of plasmid DNA versus non-plasmid DNA pieces that enter a new
cell, if they are to be stably maintained? What would be different about these DNA
elements?
4) You are working with two plasmids pRED and pWOOD. pRED confers resistance to
kanamycin (KmR), lacks an oriT, and has the lacZ gene. The pWOOD plasmid confers
ampicilin resistance (AmpR), has an oriT, and also has the same lacZ gene as pRED. You
transform E. coli (strain SLUG1, which is F+tra+lacZ-) and get KmR , AmpR, and lac+
colonies. (i) If you take a KmR and AmpR colony of E. coli strain SLUG1 and conjugate
it with E. coli strain UC1 (F-, tetracycline resistant, TcR) what would its antibiotic
resistance pattern be like? (ii) How would you explain an outcome where E. coli UC1 is
resistant to all three antibiotics (Km, Amp, Tc)? (iii) What would happen if E. coli strain
SLUG1 was F-?
5) (i) In Griffith’s experiment with Streptococcus pneumoniae smooth (S) vs. rough (R)
colonies, which form caused the mice to die of pneumonia? (ii) What happened to mice
they were inject with a mixture of heat killed S-type S. pneumoniae cells and live R-type
S. pneumoniae? (iii) What cell type did they get recover from the dead mice? (iv) How
do you explain this outcome?
