Coefficient Test
Phenol coefficient test is comparing
antimicrobial activity from chemical
components with phenol as standard
test. Diluting of desinfectant step by
step and phenol, placed in saving
sterile. Bacterium pure culture
applied as standard, added in each
tube. The test bacterium is entered in
each tube with time interval 5, 10,
and 15 minutes. All subcultures
brooded at temperature 370C during
48 hours and seen its turbidity.

 Phenol ( carbolic acid)

 Desinfectant lisol ( sample)
 Fairish steril pipette of 5 mL and 1 mL.
 Tube TSB ( Trypticase Soy Broth) or
NIOBIUM ( Nutrient Broth)
 Small and big test tube

Procedures
 Prepare microbe suspension test.
 Make standard solution of Phenol 5% ( vol/vol)
quantitative.
 Make Sample solution 5% ( vol/vol) quantitative.
 Make various concentration of phenol standard 1:80
1:90 1:100
 Make various concentration of sample 1:80 1:100
1:120 1:140 1:160 1:180 and 1:200
 Do examination of coefficient phenol according to
activity schema.
 Incubation at temperature 37°C during 48 hours.
 Observe the result of incubation and calculate the
price of its the phenol coefficient.

Example of Calculation
 The result gotten then is packed into
calculation as follows:
 Desinfectant test at diluting 1 : 450,
showing there is no growth at minute 10th
but grows at minute 5th. Diluting of phenol
at 1 : 90 showing there is no growth at
minute 10th but grows at minute5th.
 Phenol coefficient for desinfectant is :
1 : 450/1 : 90
 Phenol coefficient less than 1 indicates that
the desinfectant less effectively is compared
to phenol. Phenol coefficient more than 1
indicates that desinfectant is more
effectively than phenols.
