Each tablet contains:
Metoclopramide HCl ................................................... 10 mg
PHARMACOLOGY
DAMABEN ® which contains Metoclopramide HCl is
used in the treatment of several functional impairments
of digestive apparatus.
Metoclopramide increases the gastric motility, relaxes
the pyloric sphincter and duodenal bulb, and increases
peristalsis of the duodenum and jejunum resulting in
accelerated gastric emptying.
Metoclopramide is also endowed with an antiemetic
effect.
Metoclopramide stimulates motility of the upper gastrointestinal tract without stimulating gastric, biliary, and
pancreatic secretions.
Therefore, Metoclopramide is useful application in
numerous pathologic manifestations of the digestive
apparatus functional or following gastro duodenal ulcer,
surgical operations, anesthesia, exogenous or endogenous intoxication.
INDICATIONS
DAMABEN ® is indicated for:
- Diabetic gastroparesis.
- Metoclopramide is indicated for the relief of symptoms
associated with acute and recurrent diabetic gastroparesis.
- Nausea and vomiting.
Metoclopramide is used in the treatment of drug
related postoperative nausea and vomiting.
- Reflux oesophagitis.
Metoclopramide is indicated for the symptomatic
short-term treatment of heartburn secondary to reflux
oesophagitis.
CONTRA-INDICATIONS
- Patients with known hypersensitivity or intolerance to
Metoclopramide.
- Metoclopramide should not be used in epileptic or
patients receiving other drugs which are likely to cause
extrapyramidal reactions.
- In the presence of gastrointestinal hemorrhage, obstruction or perforation.
- Patients with “Pheochromocytoma”.
ADVERSE REACTIONS
Drowsiness, fatigue, lassitude, restlessness, constipation,
diarrhea, urticaria, dryness of the mouth, glossal or
periorbital oedema, methaemoglobinemia, extra-
pyramidal reactions such as parkinsonian symptoms,
tardive dyskinesia more likely to occur in children and
the elderly.
PRECAUTIONS
- Reduced dosages are recommended in patients with
renal failure, since the risk of extrapyramidal effects
may increase.
- Patients undergoing treatment with this drug should
not take charge of vehicles or machineries.
- It should be used with caution in nursing mothers
because Metoclopramide is excreted in breast milk.
- It should be given with caution in patients taking other
drugs that can also cause extrapyramidal reactions,
such as phenothiazines.
- Metoclopramide should not be used during the first
trimester of pregnancy. In the subsequent period of
pregnancy, the use of the drug should be limited only
in those cases of clear symptom where an alternative
intervention would not be possible, thus excluding the
frequent and simple cases of vomiting of pregnancy
and prevention of same.
DRUG INTERACTIONS
- The effects of anticolinergic drugs and narcotic
analgesics are antagonized by Metoclopramide.
- Additive sedative effects can occur when Metoclopramide is given with alcohol, sedatives, hypnotics,
narcotics or transquilizers.
- Increase absorption rate of paracetamol, tetracycline,
levodopa, ethanol, cyclosporine, and decrease
absorption rate of digoxin.
- Patients receiving MAO inhibitor therapy.
DOSAGES
Adults: 1 tablet 3 times a day. Take medication 30
minutes before meals or bedtime. All doses above may
be increased according to physician's advise.
In patients with renal impairment, the dose should be
reduced minimum 60% of normal doses.

FARMAKOLOGI
DAMABEN ® mengandung Metoklopramid HCl digunakan pada gangguan fungsi alat-alat pencernaan.
Metoklopramid meningkatkan motilitas lambung. Merelaksasi sfingter pilorus dan bulbus duodenum serta
meningkatkan peristaltik duodenum dan jejunum sehingga mempercepat pengosongan lambung.
Metoklopramid juga memiliki daya antiemetik.
Metoklopramid merangsang motilitas saluran pencernaan bagian atas tanpa merangsang sekresi lambung,
empedu dan pankreas.
Dengan demikian Metoklopramid HCl berguna untuk
pemakaian pada beberapa manifestasi patologis dari
alat-alat pencernaan baik fungsional maupun pada
ulkus gastroduodenal, pembedahan, anestesia, intoksikasi eksogen maupun endogen.
INDIKASI
DAMABEN ® diindikasikan untuk:
- Diabetik gastroparesis.
Metoklopramid diindikasikan untuk meredakan gejala
diabetik gastroparesis akut dan yang kambuh kembali.
- Mual dan muntah.
Metoklopramid digunakan untuk menanggulangi mual
dan muntah karena pengaruh obat-obatan sesudah
operasi.
- Esofagitis refluks.
Metoklopramid digunakan pada pengobatan jangka
pendek gejala rasa terbakar yang berhubungan
dengan esofagitis refluks.
KONTRA-INDIKASI
- Penderita yang hipersensitif atau tidak tahan terhadap
Metoklopramid.
- Metoklopramid tidak boleh digunakan pada penderita
epilepsi atau penderita yang memakai obat lain yang
serupa karena dapat menyebabkan gangguan
ekstrapiramidal.
- Adanya pendarahan obstruksi atau perforasi pada
saluran cerna.
- Penderita dengan “Pheochromocytoma”.
EFEK SAMPING
Mengantuk, lemah, lelah, gelisah, konstipasi, diare,
urtikaria, mulut kering, glossal atau periorbital udem,
methaemoglobinaemia, gejala ekstrapiramidal seperti
gejala parkinson, diskinesia tardive, terutama pada usia
lanjut dan anak-anak.
PERHATIAN
- Pada penderita dengan gangguan ginjal, disarankan
pengurangan dosis karena dapat meningkatkan
gejala ekstrapiramidal.
- Selama minum obat ini, jangan mengemudikan kendaraan bermotor atau menjalankan mesin.
- Hati-hati bila diberikan pada ibu menyusui, karena
Metoklopramid dieksresikan melalui air susu ibu.
- Harap diberikan dengan hati-hati pada penderita
pemakai obat-obat lain yang dapat menimbulkan
gejala ekstrapiramidal seperti obat-obat golongan
fenotiazin.
- Metoklopramid sebaiknya tidak digunakan selama
kehamilan trimester pertama. Pada periode kehamilan
selanjutnya, pemakaian dibatasi hanya pada kasus
dengan gejala yang jelas dan intervensi alternatif
tidak memungkinkan, jadi menghilangkan dan mencegah gejala mual yang sederhana dan sering pada
kehamilan.
INTERAKSI OBAT
- Bersifat antagonis terhadap kerja obat-obat golongan
antikolinergik dan analgetik narkotik.
- Menambah efek sedasi bila diberikan bersama-sama
dengan alkohol, sedatif, hipnotik, narkotik atau tranquilizers.
- Meningkatkan absorpsi dari parasetamol, tetrasiklin,
levodopa, etanol, siklosporin dan dapat menurunkan
absorpsi dari digoksin.
- Penderita yang mendapat pengobatan MAO inhibitor.
DOSIS
Dewasa: 1 tablet 3 kali seharí. Diberikan 30 menit sebelum makan atau sebelum tidur. Dosis di atas dapat
ditingkatkan sesuai dengan anjuran dokter.
Pada penderita gangguan fungsi ginjal, dosis dikurangi
minimal 60% dari dosis normal.
