Transcriptional and posttranslational heterogeneity in triple-negative breast cancer

Triple-negative carcinoma is an aggressive and heterogeneous category of breast cancer in which various
tumor suppressor genes are lost by mutation, deletion, or silencing. The diversity of triple-negative cancers
suggests that such cells might be versatile in the way that endogenous tumor suppression is subverted. Here
we report a mode of action and unusual mechanism of deactivation for growth-differentiation factor 11 (GDF11)
in triple-negative breast cancer. Using 3D cultures, intraductal xenografts, and clinical samples, we show that
GDF11 promotes a compact epithelial and anti-invasive phenotype that suppresses malignancy. Triplenegative cancers retain the GDF11 locus and expression of the protein itself. However, the bioactivity of
GDF11 is lost because of deficiencies in its convertase, proprotein convertase subtilisin/kexin type 5 (PCSK5),
causing the inactive precursor to accumulate intracellularly. Thus, triple-negative breast cancer cells harbor a
latent reservoir of GDF11 tumor suppression that can be unleashed upon restoration of convertase activity.
Sequestered GDF11 reveals a vulnerability, which may prove important given the lack of therapeutically
actionable mutations in triple-negative disease.
