Construction of episomal vectors pAnti–IGF IR and pAnti–IGF IRi. The rat insulin-like growth
factor receptor I (IGF IR) cDNA clone p118 was digested by Xho I and Kpn I, and the 0.8-kb
restriction fragment was ligated in an antisense orientation into pCEP4 (shown), yielding pAnti–
IGF IR, or into pMEP4, yielding pAnti–IGF IRi. CMV indicates cytomegalovirus immediate-early
gene enhancer/promoter; SVpA, simian virus 40 poly(A); EBNA-1, Epstein-Barr virus (EBV)–
encoded nuclear antigen 1; OriP, EBV origin of replication; Hyg, hygromycin resistance gene;
Amp, ampicillin resistance gene; Km, kanamycin resistance gene; and IGFIR, IGF IR. pAnti–IGF
IRi was constructed in a similar fashion by using pMEP4 (identical to pCEP4 but containing the
human metallothionein IIa promoter).
