STABILITY AND STORAGE RECOMMENDATIONS: When acetyl coenzyme A, [acetyl-1- 14C]- is stored at -20oC as a solid under nitrogen, the rate of decomposition is initially 3-4 % per year from date of purification. The rate of decomposition can accelerate. Stability is nonlinear and not correlated to isotope half-life. Lot to lot variation may occur. To maintain product purity, this sample should be slowly thawed at 4oC. Repeated freezing and thawing should be avoided SPECIFIC ACTIVITY RANGE: 50-62 mCi/mmol (1.85-2.30 GBq/mmol) RADIOCHEMICAL PURITY: This product was initially found to be greater than 95% when determined by the following method. It is advisable to check purity prior to use. 1. High pressure liquid chromatography on a C8 or C18 column using the following mobile phase: 50mM potassium phosphate buffer, pH 5.3 : MeOH, (85:15). 2. Paper chromatography on Whatman P-81 using the following solvent system: n-butanol : acetic acid : water, (5:2:3). CHEMICAL PURITY: Determined by ultraviolet spectrophotometry in 0.1M potassium phosphate buffer, pH 7.0. The values obtained for ratios of absorbency and λmax are within the accepted ranges. QUALITY CONTROL: The radiochemical purity of acetyl coenzyme A, [acetyl-1- 14C]- is checked at appropriate intervals using the above chromatography method.
