Material/methods: In the study, we characterize the specificity and function of virus-specific B cells at the cellular and clonal level in the children with acute EV71 infections. Results: There observed a rapid induction of virus-specific antibody-secreting B cell response in children with laboratory-confirmed EV71 infections. The frequency of IgG virus-specific antibody-secreting B cells (0.12% of total circulating lymphocytes in average) correlated significantly with the neutralization titer in the serum. A panel of monoclonal antibodies was produced from virus-specific B cells from infected children. Each of 45 EV71 capsid-specific antibodies had the uniquely rearranged VDJ pattern. 18 neutralized viruses isolated back to 1998 at least, differed in VDJ gene usages and the majority of them might recognize closely related sites around the canyon region. The interaction of missense mutations of variable regions and neutralizing/binding ability of antibodies might underline the immunological footprinting of anti-EV71 B cell response. Conclusions: The detailed analysis of EV71-specific antibody repertoire in human would further optimize the design of vaccine antigen in the future.
