Signal integration by JNK and p38 MAPK pathways in cancer development Two researchers at the CNIO - Erwin Wagner (Director of the BBVA Foundation CNIO Cancer Cell Biology Programme and Head of the Genes, Development and Disease Group) and Angel R. Nebreda (Head of the Signalling and Cell Cycle Group) have just published an important review in Nature Reviews Cancer on how molecular signalling pathways are integrated in the development of cancers. The paper describes how protein kinases of the JNK and p38 MAPK families act in a cellular context and in a cell type-specific manner to integrate signals that affect proliferation, differentiation, survival and migration. Consistent with the importance of these events in tumorigenesis, altered JNK and p38 MAPK signalling is associated with cancers in mice and humans. Studies in mouse models have been essential to better understand how these signaling proteins control cancer development and are expected to provide new strategies for the design of better therapeutic approaches. In this review the authors highlight the recent progress made in defining the functions of the JNK and p38 MAPK pathways in different cancers, as well as their potential as therapeutic targets.
