Paraffin sections:
Proteolytic enzyme digestion of deparaffi-nized section before
staining is necessary. For enhanced results it is recommended
to use Pepsin for treatment in following working protocol: =
0,1% Pepsin in 0,5 M acetic acid for 2 hours at +37°C. After
treatment slides must be well soaked with buf-fer. The antibody
dilution when used the ABC-method is 1: 10 up to 1 : 50, for 1
hour at RT.
Cryostat Sections: Type II Collagen Antibody can be used to label acetone fixed
cryostat section or cellsmears too. The optional working
dilution should be determined by the individual laboratory.
