Biosolubilisation of brown coal by yeast Bartosz Strzelecki, Natalia Kwiatos, Irena Romanowska, Hanna Miszkiewicz, Stanisław Bielecki Keywords: biosolubilisation, brown coal, yeast equipment required: flasks 500ml, tube 10 ml, centrifuge, execution time: 14 days Description: In this study is used following medium: YPG: glucose (20 g/l), yeast extract (10 g/l), peptone (20 g/l), (Agar for solid medium 20 g/l). The medium was set to pH 6,5 before autoclaving (20 min, 121oC). Microorganisms used in experiments are deposited in collection IBT. Yeast are cultivated on solid YPG medium for 7 days and then flask with 100 ml of fresh liquid YPG medium and 0.1 % (w/v) sterile powder of brown coal is inucolated sterilely. The culture is incubated for 7 days at 20oC and 180 rpm. Inoculum prepared in this way is ready to use. Biosolubilisation is conducted added 5% (w/v) of sterile pretreated brown coal and leading process for 14 days on the rotary shaker (200 rpm) at 30oC.
