Most extrahepatic tissues, such as brain, kidney, cardiac and skeletal muscles can

utilize ketone bodies, but can’t form ketone bodies. Therefore, extrahepatic
tissues are net consumers of ketone bodies.

Problem based on learning methods
1. Liver homogenate and muscle homogenate preparation: Put 3g rabbit liver and
3g rabbit muscle into two mortars respectively, make them into small pieces with
scissors. Then add 8ml normal saline respectively and grit.

2. Mark 3 tubes and add reagents according to the following table.
3. Mix, incubate at room temperature for 10min, and then filter respectively.
4. Add chromogenic agent powders into three fossae of a white porcelain plate with a spoon.
Drop 1-2 drops filtrate into fossae one by one. Observe the change of color in every fossa
and analyze the results.
Ketone bodies refer to acetoacetate, β-hydroxybutyrate, and acetone. Fatty acids, such
as butyric acid, through β-oxidation can be catabolized into acetyl CoA (coenzyme a), and

then turn into ketone bodies by ketogenesis. Ketogenesis only occurs in liver, but the
enzymes that are required by utilization of ketone bodies are absent from the liver. Therefore,
the liver is a net producer of ketone bodies.
