Summary The energy consumption of mankind and the currently available energy sources will be imbalanced in the near future if new technologies based on alternative renewable energy sources will not be developed. The most potent energy source is the Sun, and there are numerous potential energy carriers to which the energy of the Sun can be converted. These are biohydrogen, biogas, bioethanol, biodiesel e.t.c. Most of these technologies are indirect in a sense, that in the first step, the light energy is converted to agricultural products and/or wastes, these organic materials might be used for production of various energy carriers applying various fermantation technologies. The technology of the future will be based on energy carriers, which satisfy the following criteria: environmentally sound, storable, tramsportable, technologically compatible, economically feasible. Ho The actual choice depends on these parameters and might change in time. Nowadays, the bioethanol, biodiesel and biogas technology satisfy most of the criteria, therefore energy plants producing either biogas, biodiesel or bioethanol were built up worldwide. However, the cleanest energy carrier is the hydrogen, since its burning leads to the formation of pure water. Direct conversion of sunlight to biohydrogen can be principally achieved by photosynthetic microbes having hydrogen evolving enzymes, like hydrogenase or nitrogenase. However, these enzymes are ususally very sensitive to oxygen, therefore simultaneous photosynthesis and hydrogen production can not be performed by oxic phototrophic microbes. The usage of anaerobic photosynthetic bacteria might eliminate this problem, but in this case – in addition to solar energy – a sort of electron source should be available. Few purple (sulfur) photosynthetic microbes have complex hydrogen metabolism with the participation of NiFe hydrogenases and nitrogenases. They can evolve hydrogen from various organic sources, like lactate or acetete or sulfur compounds. One remarkable representative of these is Thiocapsa roseopersicina, which – as a world recorder – contains five distinct NiFe hydrogenase and at least one nitrogenase. The strain is able for direct utilization of light energy for hydrogen evolution. In the indirect biohydrogen producing strategies first, the light energy is captured by photosynthetic organisms and converted into various kinds of biomass, like energy crops, agricultural, industrial or commercial wastes. These – usually polymeric - materials of high energy contents can be converted/hydrolyzed into small units which can be used by fermentative microbes for hydrogen production. A two-stage fermentation system has been constructed to test and demonstrate the feasibility of biohydrogen generation from keratinrich biowaste. A novel aerobic Bacillus strain (Bacillus licheniformis KK1) has been isolated, which was employed to convert keratin-containing biowaste resulting in a fermentation product that is rich in amino acids, peptides. This product - supplemented with essential minerals - was metabolized by Thermococcus litoralis, an anaerobic hyperthermophilic archaeon. T. litoralis grew on the keratin hydrolysate and produced hydrogen gas as a physiological fermentation byproduct. The direct and indirect strategies are combined in the frame of EU FP6 programme. The novel approach in HYVOLUTION is based on a combined bioprocess employing thermophilic and phototrophic bacteria, to provide the highest hydrogen production efficiency. The process starts with the conversion of sugar polymer based biomass of plant origin to make a suitable feedstock for the bioprocess. The subsequent bioprocess is optimized in terms of yield and rate of hydrogen production through integrating fundamental and technological approaches. Hydrogen producing microbes are indispensable participants of biogas formation. It is shown that the biogas formation rate can be intensified by involvement of externally added hydrogen producing microbe either at mesophilic or thermophilic conditions. The enhanced biogas production could be demosntrated at laboratory and small-scale unit level. Absztrakt Az emberiség energiafelhasználása és a jelenleg rendelkezésre álló energiaforrások egyensúlya a közeljövőben fel fog borulni, ha nem fejlesztenek ki új technológiákat, melyek alternatív megújuló energiaforrásokon alapulnak. Számos olyan fontos energiahordozó létezik, melyekké - a legígéretesebb energiaforrás – a Nap energiája átalakítható. Ilyenek: a biohidrogén, biogáz, bioetanol, biodízel stb. A legtöbb technológia közvetett abban az értelemben, hogy első lépésként a fényenergia alakul át mezőgazdasági termékké vagy hulladékokká és ezek a szerves anyagok használhatók fel különböző energiahordozók előállítására megfelelő fermentációs technológiák alkalmazásával. A jövő technológiája olyan energiahordozókon alapul, melyek megfelelnek a következő szempontoknak: környezetbarát, tárolható, szállítható, technológiailag illeszthető, gazdaságilag megvalósítható. Az aktuális választást, mely az idők során változhat, ezek a paraméterek határozzák meg. Napjainkban a bioetanol, a biodízel és a biogáz technológiája a legfejlettebb, számos biogáz, biodízel és bioetanol üzemet építettek és építenek mostanában. Mindazonáltal a legtisztább energiahordozó a hidrogén, hisz égetése során csak tiszta víz képződik. A fényenergia elvben közvetlenül is hidrogénné alakítható olyan fotoszintetikus mikróbák segítségével, melyeknek vannak hidrogén fejlesztő enzimei, mint pl. hidrogenáz vagy nitrogenáz. Azonban ezek az enzimek általában igen érzékenyek oxigénre, ezért az aerob fotoszintézissel párhuzamosan hidrogén nem termeltethető. Mindazonáltal anaerob fotoszintetikus mikróbákkal ez a probléma kiküszöbölhető. Ebben az esetben azonban nap energiáján kívül valamilyen elektronforrást is kell biztosítani a hidrogén termeléshez. Számos bíbor (kén) fotoszintetikus baktériumnak komplex hidrogén metabolizmusa van, melyben NiFe hidrogenázok illetve nitrogenázok vesznek részt. Illye mikroorganizmusok különféle szerves vegyületekből, mint pl. laktát/acetát vagy redukált kénvegyületekből képesek hidrogént előállítani. Ezeknek a sejteknek egy figyelemre méltó képviselője a Thiocapsa roseopersicina BBS, mely – világrekorderként – öt NiFe hidrogenázt és legalább egy nitrogenázt tartalmaz. A közvetett biohidrogént termelő stratégiákban első lépcsőben a fényenergiát fotoszintetikus organizmusok segítségével valamilyen típusú biomasszává alakítjuk. Ilyenek lehetnek az energianövények, mezőgazdasági és háztartási hulladékok. Ezeket a magas energiatartalmú, ámde az esetek többségében polimer anyagokat először olyan egységekké kell bontani, hidrolizálni, amelyekből a fermentatív mikróbák már képesek hidrogént termelni. Egy kétlépcsős fermentációs rendszert fejlesztettünk ki keratintartalmú biológiai hulladékok biohidrogénné történő alakítására. Az általunk izolált Bacillus (Bacillus licheniformis KK1) törzs segítségével a keratintartalmú hulladékot peptidekben és aminosavakban gazdag termékké fermentáltuk. Ezt a fermentlevet – esszenciális nyomelemekkel kiegészítve – a hipertermofil archaebaktérium, Thermococcus litoralis képes volt metabolizálni és a törzs keratinhidrolizátumon nevelve jelentős mennyiségű hidrogént termelt. A direkt és indirekt közelítést kombináljuk az EU 6. keretprogram egy pályázatában. A HYVOLUTION-nak keresztelt új megközelítés egy kombinált biológiai folyamaton alapszik, melyben termofil és fotoszintetikus baktériumokat használunk a maximális hidrogén kitermelés érdekében. A folyamat növényi eredetű cukor polimerek fermentálható tápoldattá való konverziójával kezdődik. A következőkben több lépcsős integrált technológiai megközelítést alkalmazunk a hidrogén termelés folyamatának optimalizálásához. A hidrogén termelő mikroorganizmusok a biogáz képzésének elengedhetetlen komponensei. Általában a hidrogén a biogázban nincs jelen, ezért feltételezhető, hogy a hidrogénképződés sebességmeghatározó lépés. Ebből következően hidrogéntermelő mikróba adagolásával a biogáz képződésének sebessége növelhető. A megnövekedett biogáz termelést demonstráltuk laboratóriumi és félüzemi szinten mezofil és termofil körülmények között.
