Description
At least 23 different variants of Interferon-α are known. The individual proteins have
molecular masses between 19-26 kDa and consist of proteins with lengths of 156-166
and 172 amino acids. All IFN-α subtypes possess a common conserved sequence
region between amino acid positions 115-151 while the amino-terminal ends are
variable. Many IFN-α subtypes differ in their sequences at only one or two positions.
Naturally occurring variants also include proteins truncated by 10 amino acids at the
carboxy-terminal end.
Recombinant Human IFN-a 2b produced in E. coli is a single, non-glycosylated,
polypeptide chain containing 165 amino acids and having a molecular mass of 19,269
Dalton. The Interferon-α 2b gene was obtained from human leukocytes.
rHuPEG-IFN-α 2b is manufactured by attaching a 12,000 dalton methoxypolyethylene
glycol-N-hydroxysuccinimid to IFN-a 2b having a total molecular mass of 31,269 dlaton.
The rHuIFN-α 2b is purified by proprietary chromatographic techniques.
Purity
Greater than 98.0% as determined by: Analysis by SEC-HPLC using SEC column
(Tosoh, TSK GEL Alpha series, G3000PWXL column). Flow rate: 1 ml/min. Solvent:
Water. Elution time for conjugated PEG-IFN: ~5.5 min.
Endotoxin Level
Less than 0.1 ng/μg (IEU/μg) of IFN-α 2b.
Quantitation
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.924 as the extinction
coefficient for a 0.1% (1 mg/ml) solution. This value is calculated by the PC GENE
computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of IFN-α 2b as a Reference
Standard.
Specific Activity
GenScript's pegylated IFN-α 2b is fully biologically active when compared to standard.
The specific activity as determined in a viral resistance assay using bovine kidney
MDBK cells was found to be 0.7X108 IU/mg.
Storage
Pegylated IFN α-2b although stable at 15 °C for 2 weeks, should be stored at 4 °C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Formulation
Each ml contains 8 mg sodium chloride, 0.05 mg tween 80, 0.05 mg acetic acid and
2.62 mg sodium acetate pH6 and 10 mg benzyl alcohol.
Sequence analysis
The sequence of the first five N-terminal amino acids was determined and was found to
be Cys-Asp-Leu-Pro-Gln. N-terminal methionine has been completely removed
enzymatically.
