Part One - Bacterial Structures and Shapes
In this exercise, you will view prepared slides of bacteria that make capsules, spores/endospores, or
have unique shapes/structures beyond the standard rods or cocci you have seen so far. Review
lecture notes, Chapter 3 about all these microbes and structures.
Prepared Slides to View - Work In Pairs
Bacterial Capsules, 100X objective
Flagella
Fat/PHB bodies
Bacillus stained for endospores, 100X objective
Treponema or Spirochetes, unusual shape and structure, 100X objective
Clostridium
Staphylococcus
Anabaena (view at 40X) make 2 different kinds of cells - a smaller green one that is
photosynthetic and a larger colorless one that fixes nitrogen. Note and include BOTH cell types in
your final drawing.
Part Two - Slide preparation of Microbes
Procedures
Each person stains, Streptococcus, Serratia, and Yogurt - label slides as such
Mix 1 loopful of water with a loop of each bacteria and set slides aside to COMLETELY air-dry
Show your instructor your drying slides to confirm you have enough material on them
After you have finished ALL other exercises, pass dried slides through flame 3 times to heat-fix
