Iodine Test For Starch
1. Label six test tubes: 1W, 1E, 1G, 1D, 1S, and 1P
2. Add to each test tube 2ml of the following sample liquids:
a. 1W- 2ml distilled water
b. 1E- 2ml egg albumen
c. 1G- 2ml 10% glucose solution
d. 1D- 2ml carbonated soft drink
e. 1S- 2ml 10% starch solution

1P- 2ml potato juice
3. Add 0.5ml iodine solution (IKI).
4. Swirl the tube GENTLY to mix.
5. Note the final color and record below.
6. Wash out and re-use the tubes for the next part of the experiment.
Brown Paper Test for Lipids
1. Separate two areas on a piece of brown paper.
2. Label them “Water” and “Oil.”
3. Give 10 min. for the spots to dry.
4. Record which spots leave a stain (grease spot) and which do not.
Sudan IV Test for Lipids
1. In a test tube add 2ml of water and 2ml of oil.
2. Shake the tube, then let it sit for several minutes.
3. Observe and record the results.
4. Add a few drops of Sudan IV to the test tube.
5. Swirl GENTLY and let sit for several minutes.
6. Record your results.
Biuret Test for Proteins
1. Label six test tubes: 2W, 2E, 2G, 2D, 2S, and 2P
2. Add to each test tube 2ml of the following sample liquids:
a. 2W- 2ml distilled water
b. 2E- 2ml egg albumen
c. 2G- 2ml 10% glucose solution
d. 2D- 2ml carbonated soft drink
e. 2S- 2ml 10% starch solution
f.
2P- 2ml potato juice
3. Add 2ml of Biuret Reagent to each.
4. Swirl GENTLY to mix.
5. Note the final color and record below.
6. Wash out the test tubes and bring them up to the front of the lab.
