ABSTRACT Influence of Lactose Concentration on the Release of Diclofenac Sodium Formulated in Sustained Release Tablet with Ethylcellulose 10 cps as a Matrix Oki Yudiswara The purpose ofthis research was to determine concentration of lactose that can produce sustained release tablet of diclofenac sodium with ethylcellulose 10 cps as a matrix, with physical quality that appropriate to the regulations, with drug release condition of sustained release, with higher total release and achieve a zero order release. Ethylcellulose matrix tablet of diclofenac sodium using lactose as a channeling agent were prepared by wet granulation process. The paddle method was selected to perform the dissolution profiles carried out in 900 ml phosphate buffer pH 6.8. There was significant difference in drug release diclofenac sodium from matrix tablet when lactose concentration was modified from 20% to 25% and 30%. But there was no significant difference in drug release diclofenac sodium from matrix tablet when lactose concentration was modified from 25% to 30%. Release of diclofenac sodium from the matrix that contain lactose as a channeling agent was increase because lactose was easily dissolve that can increase porosity of matrix resulting in higher release of drug. The best-fit release kinetic was achieved with Higuchi's model in all formula. The release mechanism for formula 2, 3 and 4 was achieve with diffusion non-Fickian.
