RESIDENCY RESEARCH
November 1986 to December 1987: Clinical studies involving the treatment of keloids and
hypertrophic scars with intralesional bacterial collagenase and corticosteroids.
March 1987 to June 1989: Clinical studies on the ability of a topical preparation of a known
lysyl hydroxylase inhibitor to prevent the recurrence of keloids following shave excision.
January 1988 to May 1988: Restriction fragment length polymorphism patterns in the DNA
isolated from two American black patients with clinical and histopathological evidence of
dermatitis herpetiformis. Results presented at the Society of Investigative Dermatology
meeting in April 1988 and submitted for publication in the Journal of the American Academy
of Dermatology in March 1989.
May 1989 to June 1990: Use of the flash lamp-pulsed tunable dye laser for the treatment of
hypertrophic scars and keloids.
POST-DOCTORAL RESEARCH
1982 to 1984: Chilton Post-doctoral fellow in the lab of Nancy Martin, Ph.D., Assoc. Prof. of
Biochem., Div. of Molecular Biology, UT Health Sci. Ctr. (Dallas, TX). My research in this
laboratory was to develop an understanding of the regulatory process by which isopentenyl
tRNA, a nonsteroidal product of the cholesterol biosynthetic pathway, modulates HMG-CoA
reductase gene expression and cell growth. Initial studies have resulted in the production,
purification and characterization of antibodies to isopentenyl adenosine which is the
modified nucleoside found in isopentenyl tRNA. A solid phase radioimmune assay system
was developed which has been demonstrated to be the most sensitive and specific assay
method available in the published literature for detecting the presence of isopentenyl tRNA
in eukaryotic cell cytoplasm. The utility of this assay technique has been demonstrated in
collaborative studies with a lab at the Hershey Med. Ctr., Hershey, PA, where tRNA gene
4
products from cloned yeast tRNA genes were screened for the presence of isopentenyl tRNA.
GRADUATE RESEARCH ACTIVITIES
1976 to 1981: I conducted my graduate research in the lab of David Shapiro, Ph.D., Assoc.
Prof. of Biochem., U of IL (Urbana, IL) Sch. of Chem. Sci. The major theme of my research
was to characterize the molecular mechanism(s) controlling cholesterol biosynthesis in
human and rat liver cells, in vitro. HMG-CoA reductase, the key regulatory enzyme in the
cholesterol biosynthetic pathway, was purified from rat liver and used to produce
monoclonal antibodies by spleen cell-myeloma fusion methods. The resulting hybridomas
were screened for anti-reductase antibody production using a solid phase radioimmune
assay technique which I developed specifically for this system. The monoclonal antibodies
proved extremely valuable for measurements of the rate of reductase synthesis and
degradation, for the assay of HMG-CoA reductase mRNA by in vitro translation, and in the
purification of HMG-CoA reductase from gene amplification mutant cells that overproduce
HMG-CoA reductase.
UNDERGRADUATE RESEARCH ACTIVITIES
1973 to 1976: Completed 7 independent study courses under the direction of Charles
Olander, Ph.D., Dept. of Biology (Austin College). Our research investigated the in vitro
regulation of heme biosynthesis following erythropoietin stimulation of rat bone marrow
cells.
Summer 1974: Participated in a 3-month research project funded by the National Science
Foundation. The study was titled "Investigation into the Actions of Compounds that
Function as Post-Irradiation Chemotherapeutic Agents."
January 1975: Conducted an independent research project in the lab of Dr. Eugene
Goldwasser, Prof. of Biochem., Univ. of Chicago. The study included in vitro stimulation of
heme and protein synthesis by erythropoietin and examination of the effect of high levels of
3
H-thymidine on protein and mRNA synthesis in rat bone marrow cells.
Summer/Fall 1975: Worked in the laboratory of Robert Dowben, M.D., Ph.D., University of
Texas Southwestern Medical School. My research explored the differences in amino acid and
carbohydrate transport systems in lymphocytes isolated from trisomic and normal children.
1975 to 1976: Completed research for the Honors Program in Biology at Austin College with
studies on the effects of cyclic adenosine 3':5'-monophosphate on rat bone marrow
erythropoiesis, in vitro.
