OUR MISSION is to seek out and support revolutionary scientific research into the causes, treatment and prevention of all types of cancer, using cells and genes as medicine. TO THIS END we fund and monitor studies and trials that meet the highest possible standards, educate the public on the potential, and facilitate greater collaboration among the scientific and philanthropic communities to promote progress. going forward It has been a remarkable year and I want to thank our many supporters, our wonderful Board and Scientific Advisory Council members, the good people at Swim Across America, our remarkable Research Fellows who inspire us all, and everyone who contributed to the New Frontiers anniversary gala honoring Co-Founder Edward Netter. Going forward, all of us at Alliance for Cancer Gene Therapy are optimistic and whole-heartedly convinced of the potential for molecular medicine to treat and ultimately cure cancers of all types. The evidence pours in every day from our own grant recipients as well as the many other scientists throughout the country and the world who are investigating revolutionary medical alternatives. Proof positive is the recent announcement of the 2012 Nobel Prize for medicine to cell geneticists for the second year in a row. We were thrilled to see this honor bestowed on pioneers Dr. John Gurdon of the UK and Dr. Shinya Yamanaka of Japan, whose groundbreaking work paves the way to reprogramming cells. In advance of the award, Dr. Dennis O. Clegg of the University of California, Santa Barbara, enlightened guests at the gala about this remarkable therapeutic technique. Someday soon, using molecular medicine, I feel certain we will be able to reset body systems to effectively treat cancer, without horrible side effects or damage to healthy cells and genes. These scientists, like our amazing ACGT Research Fellows, are challenging old ways of thinking about the nature of disease and deploying the human genome as the blueprint to better medicine. We have been firmly committed to funding and promoting cell and gene therapies for more than eleven years and stand ever more committed going forward. Barbara Netter ACGT President and Co-Founder 1 gene therapy in the news APRIL Cancer Cell This publication of the University of Pennsylvania applauded an innovative immunotherapy approach using protein inhibitors to destroy pancreatic cancer. Most pancreatic cancers contain a gene mutation known as KRAS that encourages rapid cell growth and also cloaks the tumor with inflammatory cells to cripple the immune system. A team led by ACGT Research Fellow Dr. Robert Vonderheide disabled the tumor’s ability to make the KRAS molecule, resulting in tumor death, what he called “a house of cards effect.” J un e The Scientist The article confirmed that more and more researchers are convinced that gene therapy techniques are on the brink of common medical practice and cited groundbreaking research results published last August by ACGT Scientific Advisory Council member and Research Fellow, Dr. Carl June at the University of Pennsylvania, which described the obliteration of chronic lymphocytic leukemia cells. Dr. June’s earlier laboratory research was funded by ACGT. New England Journal of Medicine A drug developed by Bristol-Myers Squibb was proven to block a protein inhibitor to disable the shield guarding cancer and resulted in dramatic tumor shrinkages. Several major companies in the U.S. and abroad are pursuing similar drugs. A team of researchers, including ACGT Scientific Advisory Council member Dr. Lieping Chen at Yale Cancer Center, have used the technique to stimulate the immune system to destroy lung, skin and kidney cancers that had previously defied treatment. The Scientist A laboratory-grown virus was proven to be vicious to cancer while leaving healthy tissue alone, the hallmark of gene therapies. Dr. Alan Melcher led the study at the University of Leeds in the UK. This report cited several trials using oncolytic viruses that seek out and destroy tumors. “If a mutated gene is causing a problem, replace or supplement it with an accurate copy. In theory, such a strategy could not just treat, but cure countless human genetic diseases.” – The Scientist, June 2012 2 “We found that simply disabling the ability of the tumors to make this molecule leads to a house-of-cards effect that resulted in massive tumor death in experimental models.” – Robert Vonderheide, MD, D.Phil, University of Pennsylvania; ACGT Research Fellow The Wall Street Journal Foundation Medicine Inc. is developing partnerships with drug companies to use their new testing program to analyze tumors for more than 200 genes at once. In this way, doctors can more quickly and accurately match cancers to targeted therapies. Currently, analytical tools test for single gene abnormalities, and as every cancer profile requires different treatment, this requires tremendous time and exorbitant expense. 20 research institutions and 172 physicians thus far are using Foundation Medicine’s procedure and companies like Novartis, Johnson & Johnson, and Celgene have signed on. The more rapidly and accurately tumors can be profiled, the more likely a match to the best treatment will be found – another huge step forward in targeted medicine. J uly The New York Times The European Medicines Agency, equivalent to the FDA, recommended approval of a gene therapy drug developed by Dutch company UniQure that treats a rare genetic disease: lipoprotein lipase deficiency. This is the first such drug approved in the Western world, opening the door to approvals of other genetic medicines. AUGUST The New York Times Novartis and the University of Pennsylvania announced an agreement to bring to market a drug that trains a person’s immune system to kill cancer cells. Remarkable results, including complete remission, were previously reported by media sources around the world. The immunotherapy treatment was pioneered in research on chronic lymphocytic leukemia (CLL) conducted by ACGT Research Fellow, Dr. Carl June. “Now gene therapy is actually fulfilling the promise that people said it would have.” – Dr. Carl June, University of Pennsylvania, Abramson Family Cancer Research Institute; ACGT Research Fellow and Scientific Advisory Council Member “There are all these exciting and promising preclinical efficacy studies that were published in high quality journals, but they are sitting in a laboratory and not moving forward because of a lack of funding for clinical translation trials. Urbina ACGT News 2012 Grant Takes Aim at Pancreatic Cancer Herbert J. Zeh, III, MD, FACS, of the University of Pittsburgh, was named 2012 ACGT Investigator to advance research into gene therapy for pancreatic cancer. Dr. Zeh’s team deploys immunotherapy techniques to battle one of the deadliest of cancers. In laboratory research, the approach, using a viral vector, yielded 80% long-term survival rates in animal models. Dr. Zeh has utilized a similar technique for patients with brain tumors, demonstrating once again the broad application for gene therapies. The grant awards $500,000 over two to three years, culminating in a human clinical trial. 2011 Young Investigator Grant Targets Ovarian Cancer Therapy Dr. Hui Hu, PhD, Assistant Professor at the Wistar Institute, Philadelphia, PA, was awarded the Young Investigator grant to fund immunotherapy research for ovarian cancer. Dr. Hu’s study explores what is called a “quiescent state” which defines the immune system on stand-by. Cancer scientists have postulated that quiescence is not a default function, rather the T-cells are triggered to remain passive. Ovarian cancers are characterized by quiescence, inhibiting an immune system response, and this research study aims to identify novel regulatory genes and networks to battle cancer cells. The grant was made possible by the ACGT Fund for Discovery, and will provide $300,000 over three years for Dr. Hu’s innovative research. “Each day, my team and I are faced with the painful realities of pancreatic cancer, keeping us motivated to quickly unlock the mysteries of this devastating disease. But it’s only through the generosity of organizations such as ACGT that we can continue to work towards the cures. This particular grant will allow us at UPMC Cancer Center and the University of Pittsburgh School of Medicine to transition our research from the lab to the bedside, granting patients hope for longer, healthier lives.” – Herbert J. Zeh, III, MD, FACS, University of Pittsburgh Medical Center; ACGT Research Fellow New Frontiers Gala Honors Co-Founder Edward Netter Our 10th anniversary gala honored ACGT Co-Founder Edward Netter and celebrated the progress and promise of cell and gene therapy research. Event proceeds of $1.4 million will go to research, as all expenses were underwritten. Keynote speakers Robert Bazell, Chief Science and Health Correspondent, NBC News, and Dr. Dennis O. Clegg, Professor, University of California, Santa Barbara, focused their remarks on the scientific potential for targeted therapies, and ACGT Scientists enlightened attendees about the science of molecular medicine. Scientific Symposium Gathers Great Minds On the afternoon of the New Frontiers event, 30 ACGT cell and gene therapy scientists gathered to share research results and consider next steps to bring revolutionary results more rapidly to medical practice. Representing many major institutions, specialties and therapeutic approaches, ACGT Research Fellows and members of the ACGT Scientific Advisory Council shared their expectation of escalating progress in the near future. The program was moderated by Council Chair, Dr. Savio Woo, Professor and Founding Chair of Department of Gene and Cell Medicine, Mt. Sinai School of Medicine, and Margaret Cianci, ACGT Executive Director. “We believe we have provided evidence that quiescence is not just a passive default state and we are now beginning to understand the molecular mechanisms by which it happens.” – Hui Hu, PhD Assistant Professor, Wistar Institute; ACGT Research Fellow 5 Broad Strokes at Swim Across America The sixth annual swim for ACGT raised $400,000 for cancer research. Hundreds of participants boated, volunteered and cheered over 200 swimmers to a rousing finish along the Long Island Sound. Swims were also held in backyards and clubs in Southern Connecticut. Everyone is a winner at this event, with special cheers for event founding chairs John and Cindy Sites, and honorary co-chairs, Donna deVarona and John Pinto, Mary Ann Henry and Howard Rubin, Arlene and Reuben Mark, and AnneMarie and Paul Queally. ACGT-Funded Trial Yields Promising Results “In recent years, we have had exciting evidence that the vision we set out to follow so many years ago can in fact bring benefit to patients. We now face a problem engendered by this success: how to bring these biologic therapies to patients safely and expeditiously.” – Dr. Xandra O. Breakefield, Harvard Medical School, Massachusetts General Hospital; President, American Society of Gene and Cell Therapy; Member, ACGT Scientific Advisory Council Two-time grant recipient Dr. Thomas Kipps, Moore’s Cancer Center at the University of California, San Diego, reported that the trials using a combination of gene immunotherapy with chemotherapy proved a safe and more effective treatment for patients battling chronic lymphocytic leukemia [CLL] with remission rates nine times greater than those using chemotherapy alone. Research Fellow Cited for Excellence Dr. George Coukos, PhD, University of Pennsylvania, was awarded the Kaleidoscope of Hope Foundation Research Award for his gene therapy research into peritoneal cancer, as well as the Ovarian Cancer National Alliance Award for his revolutionary gene therapy treatments for ovarian cancer. Dr. Coukos is the recipient of the ACGT 2006 Dr. Judah Folkman Angiogenesis Award for Cancer Gene Therapy. Research Fellows Win Prestigious Award The Cancer Research Institute presented its prestigious William B. Coley award for distinguished research in Tumor Immunology to ACGT Fellows Dr. Carl June, University of Pennsylvania, and Dr. Michel Sadelain, Memorial Sloan-Kettering Cancer Center, for their groundbreaking work in gene therapies. Dr. June is a twotime ACGT Fellow: 2008 for his investigations into ovarian cancer and 2004 for a lymphoma and leukemia study. Dr. Sadelain received a 2004 Investigator Grant for his research into gene therapy for lymphoma and leukemia and continues to be an active voice in the research community. Both are members of the ACGT Scientific Advisory Council. Co-Founder Earns High Praise Barbara Netter, ACGT President and Co-Founder was a recipient of the YWCA Spirit of Greenwich Award for her contributions to the community, and she was named earlier this year by the Wall Street Journal as Donor of the Day.
