Abstract :
Clone recombinant of protein gene virus Infectious Bursal local isolate have been produced in
this research. Infectious Bursal Disease (IBD) virus field isolated from several part in east Java
were characterized antigenically and genotypically. Polyclonal antibody against IBD virus was
used for Agar gel precipitation test and Dot blot. Total ribonucleic acid (RNA) and messenger
RNA was isolated from bursa fabrisius 3 weeks old chicken that have been inoculated by IBD
virus. Messenger RNA was isolated from total RNA by using poly A Tract mRNA isolation system
(Promega) and synthesis cDNA by using Time Saver cDNA synthesis kit (Pharmacia).
Complementary DNA was ligated with EcoRI and then transformed to E.coli DH 5α.
Recombinant plasmid was analyzed by several restriction enzyme, SDS PAGE and
immunoblotting by specific antibody anti IBDV. Transformation in E.coli DH 5α by pUC19
produced 3 clones by cDNA encoding protein Virus Infectious Bursal.
