RTP DNA/RNA Virus Mini Kit Starting Material : serum, urine, plasma, cerebrospinal fluid, cell culture supernatants and other cell-free body fluids as well as swabs or tissue biopsies The RTP® DNA/RNA Virus Mini Kit is the ideal tool for simultaneous isolation of DNA and from up to 200 μl serum, urine, plasma, cerebrospinal fluid, cell culture supernatants and other cell-free body fluids. Specification : Amount of starting material - 200 µl serum, plasma, cell-free body fluids 1-10 mg tissue 200 µl cell culture supernatant swab time for preparation - about 35 min (incl. lysis time) Yield - depends on starting material Downstream-Application - Technology - membrane adsorption - RTP® technology Amplification qualitativ or quantitativ virus detection quantitativ RT-PCR virus diagnostic Applications : Virus RNA was isolated from 200 µl HCV infected human serum sample using the RTP® DNA/RNA Virus Mini Kit (serum contain 50.000 HCV copies/ml, detected by Cobas Amplicor Monitor® HCV Roche, Mannheim). 5 µl out of 60 µl of the eluted virus RNA was used in a in-house HCV-RT-PCR (EZ buffer/rTth). The RT-PCR was done on a GeneAmp 5700 SDS quantified against a Roboscreen Hepatitis C Virus Control Strip, coated with defined amounts of in vitro synthezised control HCV-RNA used for the generation of a standard graph (10....2500, 1000, 250 copies per tube). (Data kindly provided by Dr. Mona Amin, Roboscreen GmbH, Leipzig) Advantages : • • • • • remarkable reduction of handling steps and processing time by using preformulated and coated reagents addition of internal standards is not necessary highly efficient recovery of high-quality viral DNA and RNA from different sources, allowing very sensitive virus detection integrated Internal Controls allow the exclusion of false negative results simple storage at room temperature Procedure: Using the RTP® DNA/RNA Virus Mini Kit, the sample will be lysed in the prefilled Extraction Tubes containing preformulated lysis reagents (nonchaotropic lysis buffer, Proteinase K) and coated carrier nucleic acids as well as Internal Controls. The lysis reagents furthermore stabilize the viral nucleic acids and enhance the selective DNA/RNA adsorption to the membrane. Before the viral nucleic acids are eluted, the membranes are efficiently washed in order to completely remove all PCR inhibitors. By using the prefilled Extraction Tubes, the preparation of carrier nucleic acid solution or Proteinase K solution, as well as the pipetting of different components to the sample during the lysis, are no longer necessary. This reduces handling steps and processing time to a minimum and makes the system very simple. As a result of high extraction efficiency, the kit realizes high detection sensitivities. Purified nucleic acids are free of proteins, nucleases and other impurities and are ready to use for any downstream application, such as qualitative or quantitative virus detection.
