Description
Serum tartrate-resistant acid phosphatase exists as two isoforms (TRAcP5a
and 5b). They are derived by differential-posttranslational processing of a
common gene. Serum TRAcP5a is secreted by macrophages and dendritic
cells and increased in many patients of rheumatoid arthritis. Serum
TRAcP5b is produced from osteoclasts and elevated during bone
resorption. In addition, this antibody labels hairy leukemia cells with high
degree of sensitivity and specificity.

Immunogen
TRAcP purified from hairy cell leukemia spleen tissue
Formulation
Phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide.
Preparation
The antibody was purified by affinity chromatography.
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C.

Recommended Usage
Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the
suggested use of this reagent is 0.5 - 1.0 µg per ml. It is recommended that the reagent be
titrated for optimal performance for each application.
Application Notes
Clone 9C5 reacts with both TRAcP 5a and 5b isoforms.2 Additional reported applications (for the
relevant formats) include: immunohistochemistry of formalin-fixed paraffin-embedded sections1
and immunoprecipitation 1.

Structure
In serum, TRAcP exists as two related glycoprotein isoforms derived by differential processing of
a single gene product. Mature human TRAcP 5a isoform is a continuous polypeptide of
approximately 300 amino acids with Mw of around 33 kD. TRAcP5b contain

TRAcP is expressed intracellularly at high levels in cells of monocytic lineage, including
osteoclasts, macrophages and dendritic cells.
