They are derived by differential-posttranslational processing of a common gene. Serum TRAcP5a is secreted by macrophages and dendritic cells and increased in many patients of rheumatoid arthritis. Serum TRAcP5b is produced from osteoclasts and elevated during bone resorption. In addition, this antibody labels hairy leukemia cells with high degree of sensitivity and specificity. Reactivity Human, Cross-Reactivity: Mouse, Rat Western blot analysis of recombinant human TRACP/PAP/ACP5 protein (lane 1) and PBMC (lane 2) using anti-TRACP antibody, clone 9C5. Immunogen TRAcP purified from hairy cell leukemia spleen tissue Formulation Phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide. Preparation The antibody was purified by affinity chromatography. Storage & Handling The antibody solution should be stored undiluted between 2°C and 8°C. Application WB - Quality tested IHC, IP - Reported in the literature Recommended Usage Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application. Application Notes Clone 9C5 reacts with both TRAcP 5a and 5b isoforms.2 Additional reported applications (for the relevant formats) include: immunohistochemistry of formalin-fixed paraffin-embedded sections1 and immunoprecipitation 1. Mature human TRAcP 5a isoform is a continuous polypeptide of approximately 300 amino acids with Mw of around 33 kD.
