We have recently identified a novel bacterial genotoxin encoded by various enterobacteria species. The PhD project aims at decrypting the mechanisms by which this novel toxin leads to genotoxicity into host, and its impact on host cell genomic stability. In this context, the PhD project is centered on the bacterial mechanisms promoting intestinal genomic instability through the injection of bacterial proteins causing DNA damage into host cells. Our bacterial model is the entero-invasive pathogen Shigella. Shigella species are subgenus of E. coli strains that have acquired a specific set of genes that contribute to the Shigella pathotype. We previously showed that Shigella caused DNA damage into host enterocytic cells (1). Shigella induced DNA double strand breaks into epithelial cells, both in vitro and in vivo in the colon. Importantly, Shigella-infected cells are unable to stabilize the oncogene p53, a master regulator of the DNA repair response. The defective engagement of the p53 DNA repair in the context of a genotoxic stress constitutes a paradigm by which bacteria can predispose to genetic instability. We have recently identified the Shigella toxin causing genotoxicity (paper submitted). Therefore, the present PhD project will aim at decrypting the mechanisms by which this novel toxin leads to genotoxicity into host, and its impact on host cell genomic stability. The experimental program will document the mechanisms of cellular delivery and trafficking of the toxin into the nucleus of infected cells, and the consequences on host cell physiology, with a particular interest for the identification of mutagenic and tumorigenic potencies.
