Pre-Lab questions
1. What is the purpose of the agar?
2. What is the purpose of the incubator?
3. From which location do you think there will be the greatest amount of Bacteria? Why?
Procedure
Take a petri dish with four quadrants. Mark each quadrant with a sharpie or use the numbering that is on the petri dish. Now
as a class we will collect samples from different places around the classroom and hallways. (You must use a sterile swab and
use a new one for each area) We will place different samples in quadrants 1 & 3. For quadrant 2, use tweezers to obtain one
3-hole punch piece of paper, dip it into one disinfectant provided, and place the 3-hole punch in the center of quadrant 2.
Quadrant 4 will be the CONTROL for each dish. After the samples have been taken, record what you swabbed in each
quadrant below in the spaces provided. VERY IMPORTANT: Each petri dish must be sealed along the edges using tape.
Then put your name on the bottom plate of the petri dish. We will then place the dishes into the incubator and allow them to
grow until next Monday.

2. Which sample grew the best? Why did this sample grow more than the
others?
3. What factors lead to the rapid growth of the bacteria in the petri dishes?
4. What type of disinfectant did you choose and did it inhibit the growth of the
bacteria?
5. If you have a bacterial infection, what would a doctor prescribe you to treat the infection?
