Genes for tRNALys and two subunits of F0-ATPase

Name of Sequence. Caprine mitochondrial genes for
tRNALys, ATPase 6, and ATPase 8.
EMBL/GenBank Accession Number. X65975.
Species. Capra hircus, alpine goat.
Origin of Clone. Mitochondrial DNA (mtDNA) was
extracted from goat liver tissue, digested with EcoRI, and
cloned into pUC18. The clone with the 1,080-bp insert
containing the entire tRNALys, ATPase 8, and part of the
ATPase 6 coding region and the adjacent clone containing
the remaining part of the ATPase 6 coding region were
sequenced using a Sequenase 2.0 kit (USB, Cleveland, OH).
Both sense and antisense strands were entirely sequenced
using universal and reverse M13 sequencing primers and
five internal sequence-specific primers.
Comparison with Related Sequences. The caprine mtDNA
sequence coding for tRNALys, ATPase 8, and ATPase 6 has
83.6% similarity with the bovine (Anderson et al., 1982),
78.7% with the equine (Xu and Arnason, 1994), 77.2% with
the blue whale (Arnason and Gullberg, 1993), 75.2% with
the murine (Bibb et al., 1981), and 72.2% with the
homologous human mtDNA sequence (Anderson et al.,
1981).
Sequence Data. The tRNALys gene is flanked by the
partial cytochrome c oxidase II gene sequence at the
5′-end and the ATPase 6 gene by the partial cytochrome c
oxidase III gene sequence at the 3′-end. The deduced ATPase
8 and ATPase 6 proteins consist of 65 and 226 amino acids,
respectively.
Comments. The coding region for caprine F0-ATPase
subunit 8 is 3 nucleotides shorter than the bovine homologue. The codons for amino acids 41 and 42 (CCA ACA) in
the bovine sequence are replaced within the caprine
sequence with the single codon GCA (nucleotides 297−299).
The only difference to the universal genetic code within this
sequence is utilization of the UGA codon as a codon for
tryptophan instead of stop codon.
