Qty: 100 µL Rabbit anti-14-3-3 Protein Catalog No. 42-8300 Lot No. Rabbit anti-14-3-3 Protein FORM This affinity-purified rabbit polyclonal antibody is supplied as a 100 µL aliquot in 10 mM HEPES (pH 7.5), 150 mM NaCl containing BSA and 50% glycerol. PAD: ZMD.610 IMMUNOGEN Peptide corresponding to amino acid residues from the C-terminal region of 14-3-3 protein, conjugated to keyhole limpet hemocyanin (KLH). SPECIFICITY Specific for the ~29 kDa 14-3-3 protein. Immunolabeling of the 14-3-3 protein band is completely blocked by pre-adsorption of the antibody with the C-terminal peptide that was used to generate the antibody. REACTIVITY The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with bovine, canine, chicken, human, mouse, non-human primates, Xenopus and zebra fish based on the fact that these species have 100% homology with the amino acid sequence used as antigen. Sample Rat Western Blotting +++ (Excellent +++, Good ++, Poor +, No reactivity 0, Not applicable N/A, Not determined ND) Western blot of rat hippocampal (Hipp) lysate showing immunolabeling of the ~29 kDa 14-3-3 protein. . (42-8300 cont’d) USAGE Working concentrations for specific applications should be determined by the investigator. Appropriate concentrations will be affected by several factors, including secondary antibody affinity, antigen concentration, sensitivity of detection method, temperature and length of incubations, etc. The suitability of this antibody for applications other than those listed below has not been determined. The following concentration ranges are recommended starting points for this product. Western Blotting: 1:1000 STORAGE Store at 2-8°C for up to one month. Store at –20°C for long-term storage. Avoid repeated freezing and thawing. BACKGROUND 1 14-3-3 proteins are a family of highly conserved proteins that appear to have multiple roles in cell signaling . The proteins are abundantly expressed in the brain and have been detected in the cerebrospinal fluid of patients with different neurological disorders2. 14-3-3 proteins bind protein ligands that are typically phosphorylated on serine or threonine residues and regulate the functions of these binding partners by a number of different mechanisms3,4. The 14-3-3 proteins affect a diverse array of cellular processes including the cell cycle and transcription, signal transduction and intracellular trafficking. REFERENCES 1.
