Introduction
Growth Promotion and Cytotoxicity
Corning cellgro products undergo a variety of tests in which functionality and toxicity are
determined using growth-promotion analysis. Product performance is measured by harvest
to plant ratios or fold increase according to predetermined specifications.
Physiochemical Properties
Liquid and powder products undergo testing to determine pH and osmolality. Powders are
also tested for residual moisture. The pH and osmolality tests are performed according to
specific protocols in which all equipment is calibrated using standards which are traceable
to the National Institute of Standards and Technology.
Biological Tests
Testing of cellgro products guarantee low levels of endotoxin, mycoplasma and an absense
of contamination.
Endotoxin: Media are tested for bacterial endotoxin using the Limulus Amoebocyte Lysate
(LAL) Chromogenic Assay. For product release, each lot of standard liquid media must
have an endotoxin level of < 0.25 EU/mL. Powder products must have an endotoxin level
of < 0.25 EU/mL when reconstituted to formula weight with low-endotoxin water.
Sterility: All cellgro products are sterility tested.
Mycoplasma: Media products are tested for mycoplasma offsite using a large-volume
method described by Kern and Barile in “Isolation of Mycoplasmas from Cell Culture
by Agar and Broth Techniques,” M.F. Barile and G.J. McGarrity (1983) in Methods of
Mycoplasmology, Vol. 11,5. Razin and J.G. Tulley, eds., New York: Academic Press,
159-165.

CMRL 1066
CMRL 1066 is a modified version of Medium 199 and was designed initially for used with
L-strain cells in unsupplemented culture. It has been used to maintain monolayer growth
of permanent cell lines for extended periods of time without protein supplementation.
Although developed for use in serum-free cell culture, CMRL 1066 can be supplemented
with serum and used to support the growth of many types of cells.

Dulbecco’s Modified Eagle’s Medium (DMEM) is a modification of Basal Medium Eagle
(BME) containing four-fold concentrations of the amino acids and vitamins found in BME.
The original formulation was used to culture embryonic mouse cells and has since been
modified to support a variety of normal and transformed cells, such as primary cultures of
mouse and chicken cells.

Glasgow’s MEM is a modification of MEM formulated to support the growth of BHK-21
cells. The medium was originally based on BME with the addition of 10% tryptose

Ham’s F-10 Medium
Ham’s F-10 is a classical media designed to support the growth of a variety of cell types,
including HeLa, Chinese Hamster Overy (CHO), primary cells from tissue explants and
various types of human diploid cells. Serum supplementation may or may not be necessary
depending on the type of cell being cultured.

Iscove’s Modification of DMEM
Iscove’s Modification of DMEM (IMDM) is a highly enriched synthetic medium optimized
for rapidly proliferation cultures of high cell density. A modified form of Dulbecco’s Modified Eagle Medium (DMEM), Iscove’s contains additional amino acids, vitamins, selenium

Leibovitz’s L15 medium was originally developed without a sodium bicaronate buffer system
for use in non-CO2 equilibrated environments. It is buffered by phosphates and the amino
acid L-arginine, along with L-histidine and L-cysteine. The formation of acid metabolite
byproducts is prevented by replacing glucose with galactose and sodium pyruvate. Examples
of usage include the propagation of HEP-2, monkey kidney cells and primary explants of
embryonic and adult human tissue. Leibovitz’s L-15 supports established cell lines as well as
primary explants of embryonic and adult human tissue.

Physiochemical Properties
Basal salt solutions undergo testing to determine pH and osmolality according to specific
protocols which are in line with the National Institute of Standards and Technology.
Biological Tests
Corning cellgro prides itself on the ability to guarantee low levels of endotoxin, the absence
of mycoplasma, and an absence of contamination.
Endotoxin: Buffered salt solutions are tested for bacterial endotoxin using the Limulus Amoebocyte Lysate (LAL) Assay, following guidelines outlined by the manufacturer and the FDA
(1987). For release, each lot of buffered salt solutions must have an endotoxin level of ≤ 0.25
EU/mL. The majority of Corning cellgro’s liquid media are at or below our lowest detectable level today (≤ 0.001 EU/mL). Powder products must have an endotoxin level of
≤ 0.25 EU/mL when reconstituted to formula weight with low-endotoxin water.
Sterility: Corning cellgro selects a number of samples from each production lot to test using
the membrane filtration technique for 14 days, in accordance with USP XXIV Chapters
<71> and <1211>.
Mycoplasma: Buffered salt solution products are tested for mycoplasma off-site using
large-volume method described by Kern and Barile in “Isolation of Mycoplasmas from Cell
Culture by Agar and Broth Techniques,” M.F. Barile and G.J. McGarrity (1983) in Methods
of Mycoplasmology, Vol. 11,5. Razin and J.G. Tulley, eds., New York: Academic Press,
159-165.

Dulbecco’s Phosphate Buffered Saline (DPBS) is a buffer solution which can be used to
maintain cell culture media in the physiological pH range of 7.0 to 7.6. It can serve as an
irrigating, transporting or diluting fluid while maintaining cell tonicity and viability for a
limited period of time.

Fetal Bovine Serum (FBS) is a sterile, light brown colored liquid separated from the blood
of the bovine fetus. All material meets the approval of the U.S. Department of Agriculture
and is obtained from sources free of BSE, Foot and Mouth Disease, and other reportable
diseases pertaining to the species.
U.S. Sourced: Our U.S. sourced Premium Fetal Bovine serum, with the lowest endotoxin
and hemoglobin specifications, is collected from cattle herds only within the United States.
Also available heat-inactivated.
Human and Animal Sera
USDA: The USDA-certified Fetal Bovine Serum, also low in endotoxin and hemoglobin, is
derived from USDA-certified facilities outside the United States, primarily Mexico. Mexico
is recognized by the United States Department of Agriculture (USDA) to be free of bovine
spongiform encephalopathy (BSE) and Foot and Mouth Disease (FMD). This is also
available heat-inactivated.
Donor Calf Serum is a sterile, light brown-colored liquid collected from herds of bovine
calves located entirely in the United States. Animals are regularly inspected by licensed
veterinarians and certified to be free of diseases pertaining to the species.
Donor Horse Serum is a sterile, light brown-colored liquid collected from controlled
standing herds of animals located entirely in the United States. Animals are inspected
periodically by a licensed veterinarian. Diet and worming are strictly controlled and
documented to ensure all animals are healthy. Each lot of Donor Horse Serum is extensively
tested, and must be negative for Equine Infectious Anemia (EIA) and Equine Viral Arteritis
(EVA). Chemical constituents are measured further to ensure lot-to-lot consistency.
Porcine Serum is a sterile, light-brown liquid, collected from healthy pigs of U.S.-Origin,
and free of Foot and Mouth Disease, Rinderpest, and Bovine Spongiform Encephalopathy.
Each lot undergoes a biochemical analysis to ensure consistency.
Specialty Serum products are also offered by Corning cellgro for a variety of product
applications.
All sera is separated from the collected blood by refrigerated centrifugation, true-pooled,
filtered, and immediately frozen. The filtration process consists of a series of three 0.1
micron sterilizing filters, followed by a final “polishing” filter prior to final packaging.
Only raw serum that meets our strict specifications for endotoxin, residual hemoglobin,
pH and microbial quality is approved for production.

Introduction
Growth media is used either for cell culture or microbiological culture. These require
nutrients, supplements, buffers and other reagents specific to the product being grown to
maximize results and consistency.
There are two major types of growth media: those used for cell culture, which use specific
cell types derived from plants or animals, and microbiological culture, which are used for
growing microorganisms, such as bacteria or yeast.
Cell Culture Reagents
The most common growth media for microorganisms are nutrient broths and agar plates;
specialized media are sometimes required for microorganism and cell culture growth. Some
organisms, termed fastidious organisms, require specialized environments due to complex
nutritional requirements. Viruses, for example, are obligate intracellularparasites and require
a growth medium composed of living cells.

Trypan Blue
Trypan blue is the most common stain used in cell count and viability assays. Viable cells

Insect cell culture has become an increasingly important tool in biotechnology for the production of biologicals. The inherent growth characteristics of the insect cells make them
ideal candidates for Baculovirus Expression Vector Systems (BEVS) for the production of
recombinant genes becasue they provide advantages over both bacterial and mammalian
cultures, due to their fast doubling times and ability to grow in suspension. Insect cells
have significantly impacted protein production through the use of large-scale bioreactors to
produce recombinant proteins and products of medical and agricultural importance with high
expression levels via BEVS.
Conventional insect culture media rely heavily on animal sera and other components such as
yeast hydrolysate, lactalbumin hydrolysate, tryptose phosphate broth, and hemolymph as the
nutritional, chemical, and physiological support systems. However, the production of purified
recombinant protein has created a demand for media capable of supporting growth without
relying on these additives.
Serum-free growth media poses numerous advantages over the use of serum, including:

TAE Buffer, 10x
Tris Acetate EDTA (TAE) buffer is used in DNA agarose gel electrophoresis, both in the agarose gel itself and the running buffer. Linear, double-stranded DNA separates faster in a TAE
buffer-based system compared to Tris Borate EDTA (TBE) buffer, but the latter has a higher
buffering capacity and is thus more resistant to changes in pH. Use of TAE vs TBE buffer
depends on downstream applications. TAE is recommended for preparative electrophoresis
in which the nucleic acid bands being separated are to be used in cloning, and other work
requiring enzymatic applications.

TBE Buffer, 10x, Liquid
TBE (Tris-Borate-EDTA) has applications in DNA and RNA agarose gel electrophoresis and
polyacrylamide gel electrophoresis. TBE has a higher buffering capacity than Tris-AcetateEDTA (TAE) buffer, but the latter is more suitable for steps involving the action of enzymes
on nucleic acids because borate is an enzymatic inhibitor. Electrophoresis using TBE-containing agarose gels and TBE running buffer is much more suitable for size analysis of the
nucleic acids being separated, as TBE buffer allows these nucleic acids to better maintain
their structural integrity.

EDTA (ethylenediaminetetraacetic acid) is a polyamino carboxylic acid. It is a colorless,
water-soluble solid and is useful as a chelating agent due to its ability to “sequester” metal
ions such as Ca2+ and Fe3+. These cations are responsible for various undesirable effects in
molecular biological and cell culture processes. After being bound by EDTA, these metal ions
remain in the solution, but exhibit diminished reactivity. The use of EDTA allows for greater
control over many experiments.

SDS (Sodium Dodecyl Sulfate), 10%
Sodium Dodecyl Sulfate (SDS) is an anionic detergent with applications in SDS-PAGE
protein analysis and nucleic acid purification. It is a wetting agent that is effective in both
