Supplemental figure 1.
p53lox/lox/FAK+/+, p53lox/lox/FAKflox/+ and p53lox/lox/FAKflox/- mammary tumours were stained
for E-cadherin (upper panel) and vimentin (bottom panel). Sections were analyzed by
confocal laser scanning microscopy.
Supplemental figure 2.
Hierarchical clustering (A) and principal components analysis (B) on tumours isolated from
p53lox/lox/FAKflox/- (group A, indicated by black boxes), and p53lox/lox/FAKflox/+ mice (group B,
indicated by red colour). Histological subtype is specified by colored boxes, solid carcinoma
indicated by orange colour, carcinosarcoma by blue colour, and a biphasic tumour containing
both solid carcinoma as well as carcinosarcoma patches indicated by purple colour. When we
sampled 200 times a clustering with 2000 genes (similar to visualization) we observed that the
clusters were robust, with samples sometimes clustering with the samples from the other
cluster.

(A, C) Mammary glands from p53lox/lox/FAK+/+ (n=17), p53lox/lox/FAKflox/+ (n=15) and
p53lox/lox/FAKflox/- (n=11) mice, and p53R270H/FAKflox/+ (n=6) and p53R270H/FAKflox/- (n=15)
mice, obtained over the course of the experiment, were stained for the apoptosis marker
cleaved-Caspase 3. Image boxed in figure C indicates a cleaved caspase-3 positive cell. (B, D)
Percentage of P53lox/lox (p<0.01) and p53R270H ductal structures containing cleaved caspase-3
mammary epithelial cells. Five p53R270H/FAKflox/- mammary glands were excluded as they did
contain too few ductal structures to quantify the cleaved caspase-3 cells.

Supplemental figure 5.
Expression of selected basal-like genes in p53lox/lox/FAKflox/+ (indicated in pink) and
p53R270H/FAKflox/+ (indicated in blue) mammary tumours. Log gene expression values were
mu-sigma normalized and clustered the mice using Euclidean distance on average linkage.
We focused on selected basal-like genes, including Trp63, Krt17, Cryab, Ptn, Col17a1 and
Slug (snai2). Genes were not reordered.
