ÖZET KLİNİK ÖRNEKLERDEN İZOLE EDİLEN İNSAN CYTOMEGALOVIRUS (HCMV) SUŞLARINDA FOSFOTRANSFERAZ (UL97) VE DNA POLİMERAZ (UL54) KODLAYAN GEN BÖLGELERİNDEKİ MUTASYONLARIN VE HCMV GENOTİPLERİNİN ANALİZİ Bu araştırmada HCMV viruslarında UL54 (DNA polimeraz) ve UL97 (fosfotransferaz) genleri düzeyinde mutasyon varlığı incelenerek, izolatların gB tiplemeleri yapılmıştır. Çalışmaya HCMV infeksiyonu olan 60 olguya ait 85 örnek dahil edilmiştir. Örneklerin 64 adedinde UL54 gen bölgesi, 69 adedinde UL97 gen bölgesi 85’inde ise gB gen bölgesi PZR yöntemi ile amplifiye edilebilmiştir. PZR ürünleri UL54 için TaaI; UL97 için HaeIII ve FseI; gB için ise RsaI ve HinfI enzimleriyle kesilerek RFLP incelemeleri yapılmıştır. UL54 ve UL97 genlerine ilgili RFLP analizinde beklenenden farklı 2’şer ayrı kesim kalıbı bulunurken, bunların hiçbiri mutasyonlar için daha önce belirtilen kesim kalıplarına uyumlu bulunmamıştır. Yapılan DNA dizin analizi sonucunda; farklı RFLP kesim kalıpları elde edilen örneklerde değişen sayılarda mutasyonlar saptanmıştır. Bu mutasyonlardan birer tanesinin; UL54 analizi yapılan örneklerden sadece birinde ve UL97 analizi yapılan örneklerden gene sadece birinde bilinen aminoasit dizinlerinde değişikliklere neden olduğu tespit edilmiştir. UL54 gen dizisi temelinde yapılan filogenetik değerlendirme neticesinde iki farklı kesim kalıbı elde edilen örneklerin nükleotid dizinlerinin birbirlerinden oldukça farklı oldukları görülmüştür. gB RFLP analizi sonucunda; 57 hastanın dört farklı gB genotipinden (gB1-4; sırası ile %33.3, %28.3, %30.0, %3.3) biri ile infekte olduğu saptanmıştır. Üç hastanın (%5.0) ise birden fazla genotiple infekte olabileceği düşünülmüştür. Anahtar Kelimeler: HCMV, antiviral rezistans, UL54, UL97, gB ABSTRACT Investigation of Phosphotransferase (UL97) and DNA Polymerase (UL54) Coding Gene Regions Mutations and Genotypes in HCMV Strains Isolated from Clinical Specimens. In this study, genetic analysis of HCMV UL54, UL97 and gB genes were performed in clinically isolated HCMV strains. A total 85 plasma samples obtained from 60 previously confirmed HCMV cases were involved the study. UL54, UL97 and gB genes were amplified in 64/85, 69/85 and 85/85 samples, respectively. PCR products were digested with TaaI for UL54 region, HaeIII and FseI for UL97 region and RsaI and HinfI for gB region for restriction enzyme analysis. RFLP analysis revealed two different digestion patterns for both UL54 and UL97 regions. Hovewer, none of them were concordant to those of previously reported. Various number of point mutations were detected in samples after DNA sequence analysis performed according to the RFLP data. Deduced animoacid translations revealed that one sample with different UL54 digestion pattern and another one sample with different UL97 digestion pattern have single changes on the sequence that might lead to critical fonctional alterations on these gene products. Phylogenetical analysis based on the UL54 nucleotide sequence showed that two individual samples that have different RFLP digestion patterns are quite distinct from each other. RFLP analysis of gB region showed that, 57 patients infected with one of four different gB genotypes (gB1-4; %33.3, %28.3, %30.0 and %3.3, respectively).
